چکیده

How is it regulated? V(D)J recombination is regulated in a tissue-specific, lineage-specific and stage-specific manner. The V, D and J segments are recombined in an ordered sequence (D–J rearrangements before V–DJ). As you might expect, the process is partly regulated via the expression of RAG1 and RAG2, but it is also mediated by the differential accessibility of particular antigen receptor loci to the RAG recombinase (the RAG1–RAG2 complex). ‘Allelic exclusion’ is an additional level of regulation whereby only one allele is recombined if a productive antigen receptor is made. What’s special about the RAG genes? RAG1 and RAG2 could be remnants of an ancient transposon that inserted into the germline of a vertebrate ancestor. In vitro, the RAG proteins can excise a fragment of DNA containing RSSs and integrate it randomly into a target DNA molecule via a mechanism similar to retroviral integration and transposition. Inappropriate transposase activity of RAG1 and RAG2 could cause the DNA translocations associated with certain lymphatic tumors.

The Mechanism and Regulation of Chromosomal V(D)J Recombination

V(D)J recombination is of fundamental importance to the generation of diverse antigen receptor repertoires. We review our current understanding of the V(D)J recombination reaction and how it is regulated during lymphocyte development. We also discuss how defects in the mechanism or regulation of V(D)J recombination can lead to human disease.

V(D)J recombination intermediates and non-standard products in XRCC4-deficient cells.

V(D)J recombination assembles immunoglobulin (Ig) and T cell receptor (TCR) gene segments during lymphocyte development. Recombination is initiated by the RAG-1 and RAG-2 proteins, which introduce double-stranded DNA breaks (DSB) adjacent to the Ig and TCR gene segments. The broken ends are joined by the DSB repair machinery, which includes the XRCC4 protein. While XRCC4 is essential for both D...

Control of antigen receptor diversity through spatial regulation of V(D)J recombination.

Lymphocytes recognize a vast variety of pathogens by expressing a diverse repertoire of antigen receptor genes that are assembled by V(D)J recombination in immature B cells (Igh, Igk) and T cells (Tcrb, Tcra/d). V(D)J recombination takes place in the 3' proximal domain containing the D, J, and C gene segments, whereas 31 (Tcrb) to 200 (Igh) V genes are spread over a large region of 0.67 (Tcrb) ...

Cis-regulatory elements and epigenetic changes control genomic rearrangements of the IgH locus.

Immunoglobulin variable region exons are assembled from discontinuous variable (V), diversity (D), and joining (J) segments by the process of V(D)J recombination. V(D)J rearrangements of the immunoglobulin heavy chain (IgH) locus are tightly controlled in a tissue-specific, ordered and allele-specific manner by regulating accessibility of V, D, and J segments to the recombination activating gen...

Coding sequence composition flanking either signal element alters V(D)J recombination efficiency.

Lymphoid V(D)J rearrangement is targeted by recombination signal sequences (RSS) bordering V, D or J exons. We demonstrate that the DNA composition of flanking coding positions, particularly poly(A) or poly(T) stretches at one or both RSS, diminishes V(D)J recombination up to 100-fold. Positionally correct cleavages occur in the inhibited reactions, since the junctions formed show the same freq...

Lack of MSH2 involvement differentiates V(D)J recombination from other non-homologous end joining events

V(D)J recombination and class switch recombination are the two DNA rearrangement events used to diversify the mouse and human antibody repertoires. While their double strand breaks (DSBs) are initiated by different mechanisms, both processes use non-homologous end joining (NHEJ) in the repair phase. DNA mismatch repair elements (MSH2/MSH6) have been implicated in the repair of class switch junc...
